OD12.3 - Could not load file or assembly 'SigPlusNET.dll

While building OD 12.3 in release configuration, I get error: "Could not load file or assembly 'SigPlusNET.dll' or one of its dependencies. Access is denied". This error is produced by SGEN component. Interestingly, this error does not occur while building in Debug configuration. I am not a programmer, but I noticed that the error goes away when I turn "Generate Serialization Assembly" to off.

Has anyone encountered this error before? Will turning "Generate Serialization Assembly" to OFF, cause application to malfunction?

No, it will not cause any malfunction. It just causes it to run very slightly slower.
